SAMHD1抑制胃癌增殖及其表达调控的
机制研究
摘 要:
SAMHD1 是一种重要的细胞内核酸水解酶,在多种肿瘤中具有
抑制肿瘤细胞增殖的作用。本研究旨在探究 SAMHD1 对胃癌细
胞增殖的抑制作用及其表达调控的机制。通过采用 Western
blot、实时 PCR、细胞增殖实验以及荷尔蒙检测等方法,检测
SAMHD1 在胃癌组织中的表达量并验证其是否对细胞增殖具有
抑制作用。进一步通过 ChIP、Luciferase Reporter Gene
Assay、酵母双杂交实验等方法,分析 SAMHD1 的转录调控机
制以及参与 SAMHD1 转录调控的相关蛋白和基因。研究结果表
明,SAMHD1 在胃癌组织中的表达量明显低于正常胃组织,并
且 SAMHD1 对细胞增殖具有明显抑制作用。同时,我们发现,
miR-124-3p 和 miR-129-5p 可以通过直接结合 SAMHD1 的
3’UTR 区域来降低 SAMHD1 的表达。此外,本研究还筛选出
了这两个 microRNA 的调控基因和参与 SAMHD1 转录调控的蛋
白质。总之,本研究揭示了 SAMHD1 在胃癌中的作用及其表达
调控的机制,为胃癌治疗提供新的治疗靶点和策略。

SAMHD1 Suppresses Gastric Cancer Proliferation and Its
Mechanisms of Expression Regulation
Abstract:
SAMHD1 is an important cellular nucleic acid
hydrolytic enzyme that inhibits the proliferation of
tumor cells in many types of tumors. This study aimed
to explore the inhibitory effect of SAMHD1 on gastric
cancer cell proliferation and its mechanisms of
expression regulation. Western blot, real-time PCR,
cell proliferation assays, and hormone detection were
used to detect SAMHD1 expression levels in gastric
cancer tissues and verify its inhibitory effect on
cell proliferation. Further, ChIP, Luciferase Reporter
Gene Assay, and yeast two-hybrid experiments were used
to analyze the transcriptional regulation mechanism of
SAMHD1 and the proteins and genes involved in its
transcriptional regulation. The results showed that
SAMHD1 expression levels were significantly lower in
gastric cancer tissues than in normal gastric tissues,
and SAMHD1 had a significant inhibitory effect on cell
proliferation. We also found that miR-124-3p and miR-
129-5p could directly bind to the 3’UTR region of
SAMHD1 to reduce its expression. In addition, this
study screened the regulatory genes of these two
microRNAs and the proteins involved in SAMHD1
transcriptional regulation. In summary, this study
